ABSTRACT

A inibição da enzima colinesterase plasmática (BChE) pode ser utilizada como biomarcador para os efeitos da intoxicação por organofosforados e carbamatos. Nas aves, esta inibição ocorre de forma mais acentuada que nos mamíferos, porém poucos são os trabalhos publicados nestas espécies. O objetivo do estudo fo a dosagem da BChE em gansos-egípcios (Alopochen aegyptiacus) e nos anseriformes domésticos: gansos-domésticos (Anser anser domesticus) e marrecos (Anas platyrhynchos domesticus), para o estabelecimento de valores de referência normais. O trabalho possui ineditismo com relação à determinação desta enzima nos gansos-egípcios e domésticos. Os gansos e marrecos são mantidos em confinamento com fornecimento de alimentos e água ad libitum e em espaço adequado à sua manutenção no Instituto Adolfo Lutz (IAL), com a finalidade de fornecimento de sangue para a alimentação de triatomídeos do insetário de criação no Núcleo de Parasitoses Sistêmicas. Nos Alopochen aegyptiacus a média e o desvio padrão da BChE foram de 1.868 + 263,6 U/L, nos Anser anser domesticus 2.311 + 673,2 U/L e nos Anas platyrhynchos domesticus 4.290 + 86,11 U/L. (AU)


The inhibition of the plasma cholinesterase enzyme (BChE) can be used as a biomarker for the effects of intoxication by organophosphates and carbamates. In birds, this inhibition is more pronounced than in mammals, however there are few specific studies were conducted in this field. The aim of this study was to measure BChE in Egyptian geese (Alopochen aegyptiacus) and domestic anseriforms: domestic geese (Anser anser domesticus) and mallards (Anas platyrhynchos domesticus), not exposed to pesticides, for the establishment of normal values. The work is unprecedented regarding the determination of this enzyme in egyptian geese and domestic geese. Geese and mallards are kept in confinement with ad liditum food and water supply and in adequate space for their maintenance at the Adolfo Lutz Institute (IAL), for the purpose of supplying blood for the feeding of triatomines from the insectary of the Nucleus of Systemic Parasitoses. In Alopochen aegyptiacus the mean and standard deviation of BChE were 1,868 + 263,6 U/L, in Anser anser domesticus 2,311 + 673,2 U/L and in Anas platyrhynchos domesticus 4,290 + 86.11 U/L. (AU)

ABSTRACT Objective To establish hematotoxic alterations through clinical and paraclinical exploration in workers who are exposed to organophosphorus pesticides, carbamates and pyrethroids (OPCP) due to their work in production, packaging, distribution and fumigation processes in Cundinamarca-Colombia between 2016 and 2017. Materials and Methods A cross-sectional descriptive epidemiological study was carried out on a sample of 92 workers from six companies, mostly aged between 18 and 30 years, of which 61 % were males and 39 % females, and 71 % were workers in the operational area and 29 % in the administrative area. Univariate, bivariate and multivariate analyses were performed. Results Clinical exploration reported findings in 17 % of the sample group, of which only 2 % presented with erythrocyte cholinesterase outside the reference range. The values of hematological parameters such as peripheral blood smear (PBS) and complete blood count (CBC) were outside the range in 15 % and 47 % of the sample, respectively. Discussion The results suggest that there are hematological alterations in this group that could possibly be associated with chronic exposure to OPCP.


RESUMEN Objetivo Determinar las alteraciones hematotóxicas a través de una exploración clínica y paraclínica, en trabajadores que por su oficio se exponen a pesticidas organofosforados, carbamatos y piretroides (POCP), en procesos de producción, envase, distribución y fumigación, en el departamento de Cundinamarca Colombia. Metodología Se realizó un estudio epidemiológico descriptivo de corte transversal. Se realizó un análisis univariado, bivariado y multivariado. La muestra se conforma por 92 trabajadores de seis empresas, la mayoría entre los 18 y 30 años, de los cuales el 61 % son hombres y el 39 % mujeres, 71 % se desempeñan en el área operativa y 29 % en el área administrativa. Resultados Es de resaltar que se tienen hallazgos en la exploración clínica en el 17 % del grupo participante, solo el 2 % presenta la colinesterasa eritrocitaria por fuera del rango de referencia. Los parámetros hematológicos como el frotis de sangre periférica (FSP) presentan valores por fuera de los rangos en el 15 % y el cuadro hemático (CH) tiene valores por fuera de los rangos en el 47 % de las personas. Discusión Los resultados sugieren que existen alteraciones hematológicas en este grupo y que posiblemente podrían estar asociadas con la exposición crónica a POCP.

Malathion is an organophosphorous insecticide, used worldwide for pest and disease control; however, it could also affect the reproductive patterns of several species. The aim of this study was to determine the effects of malathion in the cellularity and sperm differentiation in testis and epididymis of rats. Twenty adult male Sprague Dawley rats were divided into a malathion-treated group (n=10, dose of 170 mg/kg via subcutaneous injection for a period of 13 days) and control group (n=10, injected only with normal saline). After treatments, the rats were sacrificed by regulated euthanasia and assessed for sperm count in testis and epididymis and epididymal teratospermia degree. The results showed a significant decrease in body, testicular and epididymal weight in animals treated with malathion. Testicular sperm counts in treated rats exhibited a significant decrease in the number of sperm compared to controls (42.56x106 vs. 95.99x106), as well as in epididymis (77.55x106 vs. 106.54x106). Concerning the degree of teratospermia, a significant increase of abnormal sperm in the epididymis of treated rats versus controls (42.1% vs. 21%, respectively) was observed. We conclude that malathion has a cytotoxic effect in rats, significantly reducing the number of sperm produced by the seminiferous tubules and affecting their quality and number during the process of maturation and capacitation in their transit through the epididymis, thus increasing the level of teratospermia.


El malatión es un insecticida organofosforado, ampliamente usado en el control de plagas y pestes, sin embargo también puede afectar a los patrones reproductivos de las especies. El objetivo de este trabajo fue determinar los efectos de malatión en la celularidad y diferenciación de espermatozoides en testículo y en epidídimo de ratas. Veinte ratas macho adultas de la cepa Sprague Dawley, fueron divididos en grupo tratado con malatión (n=10) en dosis de 170 mg/kg de peso, inyección sub cutánea (s.c.), por un período de 13 días (duración del ciclo del epitelio seminífero) y grupo control (n=10), los cuales solo fueron inyectados con suero fisiológico. Finalizado el tratamiento las ratas fueron sacrificadas por eutanasia normada y se procedió a medir el recuento espermático en testículo y epidídimo y el grado de teratospermia en epidídimo. Los resultados obtenidos muestran una disminución significativa en el peso corporal, testicular y del epidídimo de ratas machos tratados con malatión. El recuento espermático en testículo de ratas tratadas, al compararlos con ratas controles, muestra una disminución significativa en el número de espermatozoides (42,56x106 / 95,99x106), igual comportamiento se observó en epidídimo (77,55 x106 / 106,54 x106). Al determinar el grado de teratospermia se observó un aumento significativo de espermatozoides anormales, en el epidídimo de las ratas tratadas versus los controles (42,1% y 21%, respectivamente). Se concluye que malatión tiene un efecto citotóxico en ratas, disminuyendo significativamente el número de espermatozoides producidos por los túbulos seminíferos y afectando la calidad y el número de ellos durante el proceso de maduración y capacitación, en su tránsito por el epidídimo, aumentando el nivel de teratospermia.

ABSTRACT

O presente estudo descreve um surto de intoxicação por triclorfon em caprinos, que receberam pela via oral, a dosagem terapêutica do princípio ativo em questão (100mg/kg), provenientes da Universidade Estadual de Maringá, campus de Umuarama, PR. Cinquenta e três ovinos foram tratados com a mesma formulação/solução e não se observou nenhum efeito colateral nos animais referente ao tratamento com triclorfon. Das 20 cabras medicadas, cerca de 40 minutos após a administração do triclorfon, oito apresentaram os clássicos sinais clínicos de ataxia, decúbito externo-lateral, sialorreia, tremores, constrição das pupilas, dispneia com ruídos, micção e defecação involuntária, paresia espástica, timpanismo e lacrimejamento. Quase que imediatamente após a detecção destes sinais, as oito cabras foram medicadas com sulfato de atropina 1% 0,5mg/kg mais fluidoterapia. Cinco destes animais tiveram de receber nova dosagem de sulfato de atropina uma hora após a primeira aplicação, em função de alguns sinais clínicos, como tremores musculares, ainda estarem presentes. De 48 a 72 horas após a administração do triclorfon, três destes cinco animais vieram a óbito. Na necropsia, foi possível observar mucosas cianóticas, congestão de fígado, baço e rins, vasos mesentéricos congestos, vesícula biliar repleta, enfisema pulmonar, parênquima pulmonar avermelhado. Os resultados encontrados neste trabalho chamam atenção que o surto aconteceu em cabras adultas, que apresentavam boas condições clínicas e acima de tudo, receberam a dosagem terapêutica recomendada em bula pelo fabricante. Talvez isso possa indicar alguma sensibilidade mais elevada desta espécie animal à dosagem recomendada em bula do triclorfon (100mg/kg) para caprinos.


The present study describes an outbreak of trichlorfon poisoning in goats from the State University of Maringá, campus Umuarama/PR that received orally the therapeutic dose of the active ingredient in question (100mg/kg). Fifty-three sheep had been treated with the same formulation/solution and no side effects were observed in any of these sheep medicated with triclorfon. But from 20 goats medicated with trichlorfon, eight goats showed, about 40 minutes after its administration, the classic clinical signs of ataxia, external lateral decubitus, drooling, tremors, constricted pupils, noisy dyspnea, involuntary urination and defecation, spastic paresis, bloat and tearing. Almost immediately after the detection of these signals, eight goats were medicated with 1% atropine sulfate (0.5mg/kg and fluid). Five of these goats received a second dose of atropine sulfate one hour after the first application because of some clinical signs such as muscle tremor still being present. Forty-eight to 72 hours after administration of trichlorfon, three of those five goats died. At necropsy we observed cyanotic mucous membranes, congestion of liver, spleen, kidneys and mesenteric vessels, filled gallbladder, emphysema, and reddish lungs. The results of this study call attention to the outbreak that occurred in adult goats in good clinical conditions and, above all, that they received the therapeutic dosage recommended on the manufacture label. This suggests a higher sensitivity of the species to the recommended dose of trichlorfon (100mg/kg) for goats.

Se realizó un estudio comparativo y experimental para determinar el nivel plasmático de colinesterasa (con la técnica de Ensayo del reactivo de Ellman) en 51 mujeres embarazadas y 52 no embarazadas, expuestas en forma crónica a insecticidas organofosforados y carbamatos en la zona del páramo merideño y que acudieron a control y/o consulta en el Hospital tipo I de Mucuchíes (Mérida) con la finalidad de establecer la prevalencia de sus efectos en la zona mencionada y la ulterior consecuencia en el producto de la gestación. Los datos se analizaron mediante el software estadístico SPSS versión 15 para Windows. Los resultados mostraron que las pacientes embarazadas con o sin antecedentes patológicos, sus niveles de colinesterasa tienden a disminuir en cierta magnitud, se encontró diferencia estadísticamente significativa entre los niveles de colinesterasa promedio de mujeres embarazadas con antecedentes con respecto a las que no tenían antecedentes, en las mujeres no embarazadas y sin antecedentes que vivían cerca de zonas de cultivo con exposición a plaguicidas el nivel promedio de colinesterasa fue menor al nivel promedio de las mujeres en edad reproductiva no expuestas y una disminución de los valores de colinesterasa promedio en las gestantes menores de 39 años de edad. Se concluyo que las mujeres embarazadas con antecedentes patológicos presentan inhibición de niveles de colinesterasa plasmática con respeto a las no embarazadas y sin antecedentes.


A study was conducted to determine comparative and experimental plasma cholinesterase level (with the assay technique of Ellman's reagent) in 51 pregnant women and 52 non-pregnant, chronically exposed to organophosphate and carbamate wilderness area in Merida and who came to control and / or reference in the type I Mucuchíes Hospital Merida state in order to establish the prevalence of its effects on the area mentioned and further result in the product of gestation. The data were analyzed using SPSS statistical software version 15 for Windows. The results showed that pregnant patients with or without pathological history cholinesterase levels tend to decrease to some extent, statistically significant difference was found between the average cholinesterase levels of pregnant women with a history over which they had no background in nonpregnant women with no history of living near farming areas with pesticide exposure cholinesterase level was lower average to average level of reproductive age women unexposed and decreased average cholinesterase values in pregnant women under 39 years of age. It was concluded that pregnant women with medical history presented inhibition of plasma cholinesterase levels with respect to the non-pregnant and no history.

ABSTRACT

Pesticidas organofosforados são amplamente usados e seu uso constitui um grave problema de saúde pública. A ação clássica destes compostos é a inibição irreversível da acetilcolinesterase, promovendo acúmulo de acetilcolina nas sinapses e hiperestimulação colinérgica. No entanto, as consequências da exposição a baixas doses podem se estender a outros mecanismos de ação e sistemas neurotransmissores. Considerando que crianças constituem um grupo particularmente vulnerável aos efeitos de pesticidas, neste trabalho investigamos os efeitos da exposição aos organofosforados metamidofós (MET) e clorpirifós (CPF) durante o desenvolvimento sobre os sistemas colinérgico e serotoninérgico e sobre o comportamento de camundongos. Para isso, camundongos suíços foram expostos a injeções subcutâneas de MET, clorpirifós ou veículo do terceiro (PN3) ao nono (PN9) dias de vida pós-natal. As doses de exposição foram previamente escolhidas através da construção de uma curva dose-resposta que identificou como mais adequadas para este estudo as doses de 1mg/kg de MET e 3mg/kg de CPF, as quais promoveram em torno de 20% de inibição da acetilcolinesterase. Em PN10, parte dos animais foi sacrificada e foram avaliados os sistemas colinérgico e serotoninérgico no tronco encefálico e córtex cerebral. De PN60 a PN63, os animais foram submetidos a uma bateria de testes comportamentais. Em seguida, estes animais também foram sacrificados tendo sido avaliados os sistemas colinérgico e serotoninérgico. Em PN10, MET e CPF causaram alterações que sugerem aumento da atividade colinérgica respectivamente no tronco e córtex em fêmeas. No sistema serotoninérgico, apenas CPF promoveu alterações, aumentando a ligação ao receptor 5HT1A e transportador 5HT em fêmeas e diminuindo na ligação ao 5HT2. Em PN63, a atividade da acetilcolinesterase foi reestabelecida em todos os grupos. Ainda assim, MET diminuiu a atividade da colina acetiltransferase no córtex e a ligação ao transportador colinérgico.


Organophosphate pesticides are widely used and its use consist on a severe public health problem. The classic effect of these compounds involve irreversible inhibition of the enzyme acetylcholinesterase, causing an accumulation of acetylcholine at cholinergic synapses and, consequently, cholinergic hyperstimulation. However, when the doses of exposure are low, other the mechanisms of action may play a role and other neurotransmitter systems may be affected. Considering that children are particularly vulnerable to effects of these compounds, in this study we investigated the effects of methamidophos and chlorpyrifos organophosphate exposure during development on cholinergic and serotonergic systems and behavior. For this purpose, Swiss mice received subcutaneous injections of methamidophos or chlorpyrifos, or vehicle from the third to the nineth postnatal day (PN3 - PN9). Initially, a dose-response study was performed and the doses of 1mg/kg methamidophos and 3mg/kg chlorphrifos, which promoted 20% inhibition of acetylcholinesterase activity in brain were chosen to be used in the next set of experiments. At PN10, one day after exposure, a group of animals was sacrificed and the brainstem and cortex collected and stored to further analysis of cholinergic and serotonergic systems. From PN60 to PN63 the animals were submitted to behavioral tests in order to evaluate: anxiety, locomotor activity, decision making, depressive-like behavior and learning/memory. After the last test, the animals were sacrificed and the brainstem and cortex collected and stored to further analysis of cholinergic and serotonergic systems. At PN10, methamidophos and chlorpyrifos promoted alterations that suggest an increase of cholinergic activity respectively on the brainstem and cortex of females. As for the serotonergic system: only chlorpyrifos elicited alterations: There were increases in 5HT1A receptor and 5HT transporter binding in females and a decrease in 5HT2 receptor binding.

ABSTRACT

Objetivo: describir los posibles efectos adversos en salud y medio ambiente por el uso de plaguicidas en zonas productoras de Colombia e implementar las buenas prácticas agrícolas (BPA) en el cultivo de tomate. Material y método: se realizó un estudio descriptivo en el municipio de La Merced-Caldas con tres fases: una de diagnóstico, donde se seleccionaron 132 trabajadores del sector agrícola y se recolectaron muestras biológicas y ambientales. Una segunda fase de intervención en la cual se incluyeron 5 parcelas, en estas se implementaron las (BPA) y una fase de evaluación de estas parcelas. Se llevó a cabo análisis simple de las variables y se exploraron posibles asociaciones. Resultados: el tiempo de exposición a plaguicidas en promedio fue de nueve años. Con mayor frecuencia el sistema nervioso central (95,5 por ciento) fue el más afectado; seguido por órganos de los sentidos (46,2 por ciento); sistema digestivo (33,3 por ciento ); piel (21,2 por ciento) y otros (19,7 por ciento). Se encontraron niveles de organoclorados en el 97,0 por ciento (128), inhibición de la enzima acetilcolinesterasa en el 34,1 por ciento (45) de los participantes y ningún nivel del metabolito etilentiourea. En las muestras ambientales se hallaron niveles de organofosforados en tomate y suelo. En el agua y lodo se detectaron niveles de organoclorados. La producción de tomate, mostró una diferencia estadísticamente significativa entre las parcelas con BPA y las tradicionales (p=0,020). Conclusiones: se evidenciaron los riesgos por uso de plaguicidas y la necesidad de fortalecer la vigilancia sobre los potenciales efectos para la salud que pueden producir los plaguicidas y mediante el empleo de las BPA.


Objective: Describe the possible adverse effects on population health and environment due to pesticide use in agricultural zones in Colombia, and implement the best agricultural practices (BAP) in tomato production. Materials and methods: A descript intoxicative study was carried out in the municipality of Merced-Caldas, consisting in three phases: a diagnostic phase, in which environmental and biological samples were collected, using a sample of 132 agricultural workers. In a second phase, BAP were implemented, and in the third phase, the results were evaluated. A univariate analysis was completed and posible associations were explored. Results: Average length of exposure to pesticides was 9 years. The central nervous system was the most affected (95.5 percent), followed by sensory organs (46.2 percent ), the digestive system (33.3 percent ), skin (21.2 percent) and others (19.7 percent). Organoclorides were found in 97.0 percent (128), inhibition of acetylcholinesterase enzyme was found in 43.1 percent (45); no metabolite ethylenethiourea was found. In the environmental samples, presence of organophosphates was found in tomatoes and soil. In water and mud samples, organochlorides were found. There was a statistically significant difference between BPA farms and traditional farms. (p=0,020). Conclusions: Risks due to pesticide use were demonstrated, and the need to strengthen vigilance on the potential effects of pesticide use.

ABSTRACT

An outbreak of organophosphate (ORF) poisoning in cattle occurred in the central-midwestern region of Rio Grande do Sul, Brazil. The water available for 49 cattle was contaminated with ORF and carbamate (CM). Twenty cows died after a clinical course that lasted for few hours. The classical clinical signs of over stimulation of the parasympathetic nervous system were observed, including motor incoordination, aggressiveness, drooling, muscle tremors and, on occasion, diarrhea. Significant morphological changes were not observed at necropsy or at histopathological examination. ORF e CM were detected by thin layer chromatography in tissue samples of two affected cattle and in a sample from the water consumed by the affected cattle. Additionally, two other water samples accessed by the affected cattle were positive for ORF. A quantitative analysis carried out by gas chromatography in the abomasum contents of an affected cow revealed 0.93µg/g of mancozebe (CM) and 0.07µg/g of phorate (ORF).


Descreve-se um surto de intoxicação por organofosforado (ORF) em bovinos na mesorregião centro ocidental do Rio Grande do Sul. A água fornecida a 49 bovinos foi contaminada com ORF e carbamato (CM). Vinte bovinos morreram após quadro clínico de poucas horas. Foram observados os clássicos sinais clínicos de hiperestimulação do sistema parassimpático incluindo incoordenação motora, agressividade, sialorreia, tremores musculares e, em alguns casos, diarreia. Na necropsia e histopatologia não foram observadas alterações morfológicas significativas. ORF e CM foram detectados por cromatografia em camada delgada em amostras de tecido de dois bovinos afetados. Adicionalmente, uma amostra da água consumida pelos bovinos foi positiva para ORF e CM e outras duas foram positivas para ORF. Uma análise quantitativa realizada por cromatografia gasosa no conteúdo do abomaso de um bovino afetado, revelou 0,93µg/g de mancozebe (CM) e 0,07 µg/g de forato (ORF).

ABSTRACT

Clásicamente, la intoxicación aguda por órganofosforados produce una crisis colinérgica, que con frecuencia es continuada con un cuadro de debilidad muscular, expresión de un síndrome intermedio. La génesis de estos cuadros está relacionada con la inactivación de la acetilcolinesterasa por el insecticida. Mecanismos diferentes darían origen a polineuropatías y síndromes extrapiramidales tardíos. Se describe un paciente intoxicado agudamente con órganofosforados, que desarrolló una florida crisis colinérgica, que requirió ventilación mecánica invasiva. Después de tres semanas, ya recuperado de una neumonía y del síndrome colinérgico, se pudo definir un daño cognitivo de apariencia frontal, y se apreció la progresiva aparición de hipomimia, rigidez generalizada, bradikinesia y temblor, que configuraron un síndrome de Parkinson. Esta condición clínica se mantuvo al menos por dos semanas, siendo seguida de manera espontánea por una progresiva y completa mejoría del cuadro extrapiramidal y cognitivo. La literatura ha reportado, sólo por excepción casos similares, en los que se destacó tanto la aparición tardía del cuadro parkinsoniano, como su completa y espontánea remisión. Aunque la patogenia del cuadro parkinsoniano no está completamente establecida, existen evidencias experimentales que demuestran que los órganofosforados producen modificaciones en el transporte y en la recaptación de la dopamina. En este paciente se confirmó la doble acción patogénica de los órganofosforados, que habiéndose iniciado con un síndrome colinérgico agudo, finalizó con un compromiso dopaminérgico tardío. La completa recuperación de ambos efectos, permite encasillar a estos insecticidas como generadores de alteraciones funcionales, más que de gestores de daños o cambios estructurales.


Acute organophosphate poisoning usually produces a cholinergic crisis followed by muscular weakness or intermediate syndrome. The basis for these clinical manifestations is inactivation of acetylcholinesterase at the nicotinic and muscarinic nerve terminals and junctions. Different mechanism might lead to polyneuropathy and late extrapyramidal syndromes. We report a case of a male patient who ingested organophosphate with suicidal intentions. He developed a typical cholinergic crisis and required invasive mechanical ventilation. Three weeks later, frontal cognitive impairment was noticed and masklike face, generalized rigidity, bradykinesia and tremor progressively developed until a Parkinson syndrome was established. After his clinical condition had remained stable for at least two weeks, overt spontaneous improvement in motor and cognitive functions was observed. Similar reports in literature are infrequent. Although the pathophysiology that underlies extrapyramidal manifestations due organophosphate poisoning remains unclear, experimental evidence demostróte that organophosphate compounds impair dopamine transport and uptake. This case report suggests that organophosphate might act through a double pathogenic action i.e an initial acute cholinergic syndrome and a delayed disfunction in dopaminergic pathways. Complete spontaneous resolution of both effects allow us to classify organophosphate substances as a cause of functional impairment in the basal ganglia.

Un estudio descriptivo sobre las condiciones de salud y su relación con la exposición a plaguicidas, de los “fumigadores” de la campaña contra el dengue en el estado Aragua, fue realizado en una muestra no probabilística de 50 trabajadores, a los cuales se les aplicó una encuesta para investigar: características personales y de trabajo, conocimientos sobre insecticidas y enfermedades asociadas, además de inspeccionar locales, equipos y condiciones de trabajo. De la muestra general, 28% resultaron fumadores. Los grados de instrucción predominantes fueron primaria y secundaria (44%). El promedio de años como fumigador fue de 14,3. Sólo 40% tiene conocimiento de enfermedades producidas por organofosforados, 62% presentó antecedentes de intoxicación. 30% refirieron presentar enfermedades cardiovasculares, gastrointestinales y neurológicas. A mayor cantidad de años de labor, mayor la intensidad de signos y síntomas llegando a producir enfermedades. 38% de los niveles de Acetilcolinesterasa medidos, resultaron alterados. Las variables: grupo de edad, grado de instrucción, fumar, años de labor, falta de conocimiento de enfermedades y antecedentes de intoxicación, se asociaron a valores anormales de colinesterasa, sin presentar significancia estadística (P>0,05). La selección de los trabajadores, su capacitación, dotación de equipos y materiales adecuados, su vigilancia clínica epidemiológica, su evaluación permanente y su respectivo tratamiento deben ser revisados y tenidos en cuenta por la Corporación de Salud del estado Aragua (CORPOSALUD), haciéndolas, normas de obligatorio cumplimiento


A descriptive study on the conditions of health and their relation to the exhibition to pesticides, and of the “fumigators” of the campaign against dengue in the Aragua state, sampled 50 workers, to whom a survey was applied to investigate: personal characteristics of work, knowledge on insecticides and diseases, besides an inspection the premises, equipment and work environment. From this general sample 28% were smokers. The predominant educational level was elementary school and high school (44%). The fumigator average of working years was of 14.3. Only 40% had knowledge of diseases produced by organophosphate, 62% presented poisoning antecedents. 30% claimed to display cardiovascular, gastrointestinal and neurological diseases. The greater the number of work years, the greater the intensity of signs and symptoms of associated diseases. 38% of the measured levels of acetylcholinesterase were altered. The variables: smoking, years of work, educational level, age group and the lack of knowledge of diseases and intoxication antecedents were associated with abnormal values of acetylcholinesterase, without showing statistical significance. The selection of the workers, their qualification and suitable equipment and materials, their clinical epidemiologic monitoring, their permanent evaluation and respective treatment must be reviewed and taken into account by CORPOSALUD (Corporación de Salud del estado Aragua), making them norms of obligatory fulfillment

Diseño Metodologico Tipo de Estudio: Corresponde a una investigación analítica, del cumplimiento de normas de manejo de pacientes intoxicados por plaguicidas, principalmente por Fosfina, Organofosforados, Paraquat y Rodenticidas. El Universo esta constituido por 46 pacientes correspondientes al total de ingresos con diagnóstico de intoxicación por estos plaguicidas, por el servicio de Emergencias del Hospital “Roberto Calderón Gutiérrez.” de 2008 a 2009. Resultados: Se recopilaron 46 casos de intoxicaciones por plaguicidas Fosfina, Paraquat, Organofosforados y Rodenticidas con predominio del sexo masculino, de escolaridad primaria, con una edad media entre 15 y 25 años. Geográficamente, la mayor proporción de casos corresponde a la zona urbana, sin ocupacion y solteros. El plaguicida mayor utilizado es la Fosfina, con una severidad global del 60% y una mortalidad general del 72%. La mayoria con finalidad autolítica, utilizando la vía digestiva como via de ingreso del tóxico...

ABSTRACT

Os agrotóxicos são substâncias usadas para exterminar pragas ou doenças quecausam danos às plantações. Os organofosforados são agrotóxicos responsáveis por 70 por cento das intoxicações ocupacionais e atuam inibindo a enzima acetilcolinesterase por meio da fosforilação do aminoácido serina do sítio ativo. Existem diversos métodos de diagnóstico para monitorar a exposição a estes compostos, tais como: identificação e análise quantitativa de compostos livres noplasma; identificação e análise quantitativa de metabólitos na urina; determinação da atividade de colinesterases no plasma ou no sangue total. Entretanto, estas metodologias possuem algumas limitações, como por exemplo, a falta de especificidade, tecnologias de análise caras e variações interindividuais (sexo, idade, etc.). Uma tentativa de contornar estas limitações na quantificação da atividade de colinesterases é o desenvolvimento de metodologias baseadas na detecção da enzima fosforilada através de técnicas imunoquímicas. Este estudo teve como objetivo desenvolver um método de imunoensaio para o diagnósticorápido da exposição humana aos organofosforados pela detecção deacetilcolinesterase inibida no sangue humano por anticorpos aviários produzidos contra uma sequência de aminoácidos ao redor da serina fosforilada do centro catalítico da enzima. Para isto, foram adquiridos dois decapeptídeos sintéticos correspondentes ao sítio catalítico "não fosforilado" (SOH) e outro com o resíduo de serina fosforilado (SOP). Estes peptídeos foram conjugados com albumina bovina de soro (BSA), injetados em galinhas poedeiras da raça Isa-Brown e anticorpos IgY foram obtidas das gemas dos ovos das galinhas imunizadas. A especificidade dos anticorpos IgY anti-SOH e anti-SOP foi testada por meio de métodos ELISA constatando-se que estes anticorpos reconhecem os peptídeos livres, os peptídeos conjugados com BSA e a própria BSA...

Todos os agrupamentos humanos que se organizam para o trabalho usam rios, lagos ou lagoas como depósitos para a decomposição de matéria indesejável. A contaminação do meio aquático por herbicidas e agrotóxicos derivados de práticas agrícolas se tornou, faz tempo, um problema de importância mundial. Precisamos de informações detalhadas sobre a bioquímica da intoxicação de peixes nativos para avaliarmos quais os efeitos de agrotóxicos sobre os processos bioquímicos que mantêm o ciclo de vida dos peixes em águas do Brasil. Organofosfatos, que são agrotóxicos de uso disseminado, podem interargir com as B-esterases butirilcolinesterase (EC 3.1.1.8) e carboxilesterase (EC 3.1.1.1) presentes no fígado e no plasma. Tanto a butirilcolinesterase (BChE) como a carboxilesterase (CarbE), se presentes em concentrações relativamente elevadas, agem como limpadores estequiométricos ("scavengers" moleculares) por ligarem o átomo de fósforo do grupo P=O com a hidroxila de uma serina presente nos seus sítios ativos. Em nossos resultados observamos que curimbatá possui a CarbE plasmática (IC50 74 nM) mais sensível ao organofosfato metilparaoxon quando comparado ao pacu (IC50 691 nM). Isolamos CarbE dos plasmas de curimbatá e pacu. Piavassu não possui uma atividade expressiva de CarbE no sangue, por isso não a isolamos. O tipo e a distribuição das esterases nos tecidos são particulares da espécie. Curimbatá tem alta atividade de CarbE no fígado (237,8 U.g-1) e no sangue (29,85 U.mL-1), pacu é dotado de alta atividade de BChE (134,0 U.g-1) e CarbE (149,6 U.g-1) no fígado, mas o piavussu conta apenas com a BChE do sangue (17,87 U.g-1). Este arsenal enzimático foi suficiente para proteger as AChE de cérebro, músculo e coração das três espécies e evitar a sua intoxicação leve por 0,2 mg metilparation/L. A abordagem cinético-bioquímica para conhecer a inibição das esterases presentes nos tecidos de diferentes espécies de peixes por agrotóxicos é uma ferramenta útil...


Every human group who organizes to work together uses rivers, lakes or ponds as places in which undesirable substances are deposited for decomposition. Contamination of the aquatic environment with herbicides and pesticides derived from agrucultural practices has become a problem of global importance since a long ago. We need detailed information on the biochemistry of the poisoning of native fish to assess the effects of pesticides on the biochemical processes that maintain fishes' life cycle in waters of Brazil. Organophosphates, which are widely used pesticides, can interact with the B-esterases butyrylcholinesterase (EC 3.1.1.8) and carboxylesterase (EC 3.1.1.1) in plasma and liver. Butyrylcholinesterase (BChE) and carboxylesterase (CarbE) in relatively high concentrations act as stoichiometric scavengers by linking the phosphorus atom of the P=O group with the serine's hydroxyl they have in their active site. Our results show that the CarbE of curimbata plasma (IC50 74 nm) is more sensitive to the organophosphate metilparaoxon than CarbE of pacu plasma (IC50 691 nm). We isolated CarbE from curimbata and pacu's plasma. Piavussu does not have an expressive activity of CarbE in plasma, so we did not isolate it. The type and distribution of esterases in tissues are peculiar to a species. Curimbata has high CarbE activity in the liver (237.8 U.g-1) and blood (29.85 U.mL-1), pacu is equipped with high activities of BChE (134.0 U.g-1) and CarbE (149.6 U.g-1) in the liver and piavussu relies only on BChE of blood (17.87 U.g-1). This enzymatic arsenal was sufficient to protect AChE from brain, muscle and heart of the three species and protect them against mild intoxication by methilparathion (0.2 mg/L). The biochemical kinetic approach that allows understanding of the inhibition of the esterases in tissues of different fish species is a good tool capable of anticipating the harmful consequences of these drugs.

Os agrotóxicos são amplamente utilizados no meio rural e sua larga utilização, desprovida de medidas de biossegurança apropriadas, vem provocando impactos potenciais tanto para a degradação ambiental quanto para o organismo humano, aumentando a incidência de intoxicações ocupacionais. O objetivo do presente estudo foi revisar e discutir a respeito da exposição aos agrotóxicos e sua influência na função e estrutura do sistema auditivo. Foram revisados periódicos nacionais e internacionais relevantes indexados no PubMed, Medline, Bireme e LILACS, assim como capítulos de livros e dissertações publicados no período entre 1987 e 2009. As pesquisas sugerem que a exposição, principalmente crônica, aos agrotóxicos induz a alterações funcionais no sistema vestibulococlear, manifestando ainda uma série de sinais e sintomas. Apesar da escassez de dados sobre a influência do organofosforado nas características morfológicas do aparelho auditivo, os estudos indicam que a exposição de caráter agudo aos agrotóxicos pode também ser responsável por alterações no sistema auditivo, caracterizadas por lesões estruturais na cóclea, principalmente nas células ciliadas externas.


Pesticides are widely used in agriculture, and the widespread application of these agents, without the appropriate use of bio-security measures, has been causing potential impacts for environmental degradation, as well as for the human organism, increasing the incidence of occupational poisoning. The aim of the present study was to review the available literature and to carry out a discussion regarding pesticides exposure and its harmful effects on the auditory system's function and structure. Relevant national and international journals indexed in PubMed, Medline, Bireme and LILACS were reviewed, as well as book chapters and dissertations published between 1987 and 2009. The available data suggest that chronic exposure to pesticides lead to functional alterations in the vestibulocochlear system, as well as a series of signs and symptoms. In spite of the scarce data describing organophosphorus influence on morphological characteristics of the auditory system, the literature suggest that acute exposure to pesticides can also be responsible for auditory system alterations, characterized by structural lesions in the cochlea, mainly in outer hair cells.

ABSTRACT

Los plaguicidas organofosforados (OP) son masivamente aplicados en el Alto Valle de Río Negro y Neuquén, afectando al ecosistema. Utilizamos un modelo embrionario de anfibios (Rhinella arenarum) para estudiar mecanismos por los cuales OP como metilazinfos (MA) y clorpirifos (CP) podrían provocar teratogénesis. Los embriones fueron desarrollados en diferentes concentraciones de MA o CP hasta opérculo completo (OC), analizando: malformaciones, histología, glutatión reducido (GSH) y enzimas antioxidantes, poliaminas, actividad de ornitina-decarboxilasa (ODC) y proteínaquinasa-C (PKC). Ambos OP provocaron un incremento tiempo/concentración-dependiente de malformaciones, llegando a 100% de teratogénesis en estadios avanzados y a las mayores concentraciones, incluyendo: exogastrulación, curvaturas de aleta caudal, acortamiento axial, edema, y atrofia branquial. Se evidenció una condición de estrés oxidativo creciente: las enzimas GSH-dependientes (S-transferasa (GST), peroxidasa y reductasa) fueron inducidas tempranamente a bajas concentraciones, pero inhibidas en elestadio de OC a altas concentraciones, junto con una caída significativa de GSH (62%) para MA. MA incrementó significativamente (18X) la actividad de ODC en OC, aumentando los niveles de putrescina (60%) pero disminuyendo espermidina (56%) y espermina (100%); CP disminuyó en estadios tempranos la actividad de ODC y niveles de poliaminas. La disminución de poliaminas podría deberse al incremento de degradación por poliamino-oxidasa, contribuyendo al estrés oxidativo inducido por OP. Esto causaría la disminución de GSH, y la activación de PKC en OC (55%), que participaría en el control positivo de GST y ODC. Finalmente, el estrés oxidativo y la disminución en los niveles de poliaminas podrían ser causantes de alteraciones del desarrollo embrionario.


Organophosphate (OP) pesticides are widely applied in the region of Alto Valle de Río Negro y Neuquén, affecting the ecosystem. We use an amphibian embryonic model (Rhinella arenarum) in order to assess the mechanisms by which the OP pesticides azinphos methyl (AM) and chlorpyrifos (CP) could cause teratogenesis. The embryos were developed in different concentrations of AM or CP until they reached the stage of complete operculum (CO). We analyzed malformations, histology, reduced gluthatione content (GSH) and activity of antioxidant enzymes, polyamine content, ornithine decarboxilase (ODC) and protein kinase C (PKC) activities. Both OP pesticides caused a time-and dose-dependent increase in the number of malformations, reaching 100% teratogenesis in late embryonic development at the highest OP concentrations used. Malformations assessed include exogastrulation, caudalfin curvature, axial shortening, edema, and gill atrophy. Increasing evidence of oxidative stress was observed: GSH dependent enzymes (S- transferase, GST; peroxidase and reductase) were early induced in embryos exposed to low concentrations of the OP pesticides, but their activities were inhibited in the stage of CO at high concentrations of OP. These changes were accompanied by a significant decrease in GSH content (62%) in embryos exposed to AM. Besides, AM significantly increased (18X) ODC activity in the stage of CO, along with putrescine levels (60% of increase) but spermidine and spermine levels were significantly decreased (56% and 100%, respectively). The OP pesticide CP caused and early decrease in ODC activity and polyamine levels.The decrease in polyamine levels could be due to an increase in their degradation by polyamine oxidase, contributing to the oxidative stress induced by OP. This, in turn, would cause the decline in GSH levels and the activation of PKC in the embryonic stage of CO (55%), which is involved in the positive feedback of GST and ODC...

ABSTRACT

This review briefly considers the testicular damage elicited by environmental chemical pollution. It includes a short comment on environmental toxicology as an introduction to environmental chemical pollution, highlighting the importance of this current field of study and its impact on male reproductive health. Furthermore an experimental animal model addressing the effect of organophosphorated agropesticides as a testicular toxicant is presented. Moreover two relevant chemical contaminants and their effect on the testis, such as the classical case of lead and the rarely reported case of Boron on spermatogenesis, are considered. Additionally, the subject of biosentinel species and their relevance for the monitoring of pollution in aquatic and/or terrestrial ecosystems is considered. In conclusion, it should be stressed that environmental health is closely related to the reproductive health of all living beings.


Esta revisión considera el daño testicular provocado por la contaminación química ambiental. Incluye un breve comentario sobre toxicología ambiental a modo de introducción respecto a la polución química ambiental y destaca la importancia de este campo de estudio actual y su impacto sobre la salud reproductiva masculina. Además se presenta un modelo experimental animal concerniente al efecto de agropesticidas organofosforados como tóxicos testiculares. Se consideran dos contaminantes químicos relevantes y su efecto en el testículo como son el clásico caso del plomo y el menos conocido caso del boro y sus efectos sobre la espermatogénesis. También se trata el tema de las especies biocentinelas y su importancia para el monitoreo de la evolución de ecosistemas acuáticos y/o terrestres. En conclusión, es necesario insistir que la salud medioambiental está íntimamente relacionada con la salud reproductiva de todos los seres vivos.

ABSTRACT

Los agropesticidas organofosforados (OF) son ampliamente usados para incrementar la producción alimentaría. Sin embargo, se ha demostrado que inducen alteraciones a nivel testicular, relacionadas con la disminución de la fertilidad tanto en humanos como en animales. Por otra parte, diferentes estudios han sido llevados a cabo para desarrollar compuestos químicos o naturales que puedan inducir un efecto antagónico sobre los OF. En estudios anteriores, un extracto de una planta de los Andes peruanos (Maca) ha sido reconocido por su acción estimulante sobre la espermatogénesis. En el presente trabajo se estudiaron los efectos de ambos agentes externos sobre el testículo. Una población de 52 ratones machos adultos de la cepa CF1 fue dividida al azar en 4 grupos (Control, Malation, Maca, Malation-Maca), con intervalos de sacrificio de 1, 7, 14 y 21 días. Para analizar el grado de alteración inducida por estos agentes, se utilizaron técnicas de histomorfometría con ayuda del programa Image tools 3.1, en secciones testiculares. Nuestros resultados muestran que la altura del epitelio aumentó al día 1 en el grupo tratado con malation, cayendo al día 7 y llegando a valores similares al control, al día 14. Sin embargo, el grupo Malation-Maca no mostró cambios significativos. El lumen tubular, disminuyó al día 7 y 14, para normalizarse al día 21 en el grupo Malation. Sin embargo, en el grupo Malation-Maca el lumen tubular sólo bajó al día 14. El diámetro tubular disminuyó a los días 7 (p < 0.01), 14 y 21 en el grupo Malation, con respecto al control (p< 0.05). Sin embargo, el grupo Malation-Maca mostró valores normales. En conclusión, es posible establecer que el daño inducido por Malation es revertido al día 21 post administración de maca.


Organophosphoric (OP) agropesticidas are amply used to increase food production. However, it has been verified that they induce alterations at testicular level related to the diminution of fertility in humans as in animals. On the other hand, different studies have been made to develop chemical or natural compounds that can induce an antagonistic effect to OP. In previous studies an extract from a plant, from the Peruvian Andes (Maca) has been recognized by its stimulating action on spermatogenesis. In the present study the effects of both external agents were evaluated on testicular sections of testis of adult male mice on a population of 52 mice CF1, divided at random in 4 groups (Control, Malathion, Maca, Malathion-Maca), with sacrifice intervals of 1,7, 14 and 21 days. By means of morphometric technique and using the "Image Tools 3,1" software, the histology of testicular sections was evaluated, to analyze the degree of alteration induced by these agents. The epithelial height mainly has a rise in day 1, for Malathion group and then fall to day 7 to be normal by day 14. However, Malathion-Maca groups show no changes. The tubular lumen decreases at day 7 and day 14 to be normal by day 21 in Malathion group. However, in Malathion - Maca group, the tubular lumen decreases only at 14 days. The tubular diameter, at day 7 (p<0.01), 14 and 21 in Malathion group decreases respect to control (p<0.05), However, Malathion-Maca groups show normal values. In conclusion, it is possible to establish that the damage induced by Malathion is reverted by 21 days post-administration of Maca.

ABSTRACT

La prevención del daño por exposición acontaminantes ambientales requiere identificar biomarcadores que representen una medida del proceso toxicológico antes de la manifestación de daños irreversibles. Considerando la alta vulnerabilidad del período de gestación, este trabajo fue diseñadopara evaluar el impacto de la exposición in utero a plaguicidas organofosforados (OP) y la utilidad de biomarcadores de exposición/efecto en tejidos fácilmente accesibles. Se entrevistaron 342 embarazadas del área frutícola del Alto Valle de Río Negro y Neuquén donde se alternan períodos de 6 meses de fumigación y receso. Se registraron características, hábitos y lugar de residencia materna, parámetros del neonato y de la placenta a término. Los estudios analíticos incluyeron colinesterasas sanguíneas, hemograma de las embarazadas y enzimas placentarias. Los datos fueron analizados por ANOVA regresión y correlación. La actividad de colinesterasa plasmática en época de fumigación disminuyó significativamente (18 %) y presentó asociacióncon la duración de la exposición. Tanto la actividad de acetilcolinesterasa (AChE) como catalasa (CAT) placentariaaumentaron en dicho período (40%) y la edad gestacional disminuyó en dos semanas. El perímetro cefálico aumentó por la residencia rural materna y por la fumigación en el hogar. Este parámetro se asoció con la actividad de CAT. Estos resultados indican que el uso combinado de la AChE y CAT placentarias, enzimas involucradas en eventos adaptativos, serían herramientas útiles en programas de monitoreo ambiental. Asimismo, corroboran que este tejido, el más accesible de la tríada madre – placenta– feto, muestra efectos acumulativos que reflejan el ambiente intrauterino.


Improvement of children’s health requires preventing environmental exposures and identifying predictive biomarkers that represent the toxicological process before irreversibleinjury appears. Considering the high vulnerability of the intrauterine development, this study was designed to investigate the impact of in utero exposure to organophosphate pesticides and biomarkers of early effects in easily asequible tissues. A cohort of 342 women living in the agricultural area of High Valley of Río Negro and Neuquén, where 6 month spraying and non spraying periods are alternated, were interviewed in order to characterize their demographic characteristics and lifestyle habits. Morphometric information about the offspring and placentas were recorded. The analytical studies included pregnant womenhemogram, blood cholinesterases as well as placental enzymes activities. Data were analyzed by ANOVA, regression and correlation. During spraying season plasma cholinesterase diminished significantly (18%) and was associated to exposure length. Inthis period, placental acetylcholinesterase (AChE) and catalase (CAT) increased in a similar extent (40%) and gestational agediminished in two weeks. An increase in cephalic circumference was observed in newborns from women residents in rural areas and also in ones having indoors fumigation habits. This parameter was associated to CAT activity. These results indicate that the combined use of the placental AChE and CAT, enzymes involved in adaptative events, seems to be a useful tool in environmentalmonitoring programmes. Also, they corroborate that this tissue, the most accessible and available component of the triad mother-infant-placenta shows cumulative effects that reflect the intrauterine environment in which the newborn was developed.
